The Effects of Thyroid Gland on Langerhan¢¥s Islet of Pancreas and Carbohydrate Metabolism in Liver and Muscle


Abstract
A histochemical study has been made to observe the effects of thyroid gland on Langerhan¢¥s islet in pancreas and carbohydrate metabolism in liver and muscle.
The rabbits employed in this experiment were all male, weighing 11.7 to 2.0 Kg ; devided into three groups of normal, hyperthyroidism and hypothyroidism.
The first group of normal rabbits were fed with ordinary diet. For producing hypothyroidism in the second group, thyroidectomy was performed, and the hyperthyroidism in the third group was produced by _ i oral, daily administration of 0,2 gm of thyroid Strong(Park, Davis & Co.) per Kg. of body weight for two ¢¥ weeks.
Thyroidectomized rabbits in the second group were killed at intervals of 1, 3, 7, 10, 20 days and 1, 2, and 3 months after the operation and the tissues excised from pancreas, liver, and skeletal muscle of sacrificed animal within the heart beats were still present, and were fixed in absolute alcohol and Zenker¢¥s solution.
The third group of rabbits which had been induced hyperthyroidism were killed and tissues from pancreas, liver and skeletal muscle were fixed as in the second group.
For the purpose of microscopic examination, Periodic-acid Schiff stain was applied for the demonstration of glycogen granules. And for the secretory granules in Langerhan¢¥s islet cells, the Mallory-Azan stain ¢¥ was empleyed.
In addition to these special stain, Haematoxylin-Eosin stain was also applied for the general features of the tissue.
A urine test by Benedict¢¥s method was performed to observe the occurance of glucosuria, if any, in both group during the whole course of the experiment.
The histochemical aspects in result of above experiment were as follows.
A. Hyperthyroid group.
1) Slight decrease of secretory granules in the B-cell were observed.
2) Marked depiction of liver glycogen,granules in ,the peripheral.-portion of lobules were noticed i
3) No¢¥marked changes were seen from the skeletal muscle by both of P.A.S. and H. & E. stain.
B. Thyroidectomized group.
1) Up to the "one month after" the thyroidectomy, no .definite histochemical changes were evident. However, in animals of 2 to 3¢¥months duration, an increase in minute degree of secretory _granules of B-cells were observed. from the pancreas.
2) During the first 10 days after the thyroidectomy, no inteiise differences were seen from that of normal group. But in animals of one to three months duration there was moderate or marked increase of liver glycogen.
3) No marked changes were observed from the muscle by the examination of P.A.S. and H. & E. stain.
C. Glucosuria were negative in the both of thyroidectomized and hyperthyroid groups during the whole course of the experiment.
